What Is MOTS-c?

MOTS-c is a short peptide encoded within mitochondrial DNA — making it one of the only known peptides of mitochondrial (rather than nuclear) origin. Its full name, Mitochondrial Open Reading Frame of the 12S rRNA-c, reflects where it is encoded in the genome: within the 12S ribosomal RNA gene of the mitochondrial chromosome.

First characterised in 2015, MOTS-c has rapidly become one of the most actively investigated compounds in metabolic and healthy lifespan pathway research. It functions as an exercise-responsive mitochondrial signalling peptide — meaning its levels naturally rise in response to physical activity — and has been shown to regulate metabolic homeostasis, insulin sensitivity, glucose utilisation, fat oxidation, and inflammatory burden through a mechanism that operates from the mitochondria outward to the cell nucleus and systemic circulation.

Because of its metabolic and exercise-mimetic properties, MOTS-c is of particular interest for research into obesity, insulin resistance, age-related metabolic decline, and physical performance — especially in populations where the metabolic benefits of exercise are reduced or inaccessible.


How It Works — Mechanism of Action
