What Is IGF-1 LR3?

IGF-1 LR3 is a synthetic analogue of human Insulin-like Growth Factor-1 (IGF-1), engineered with two specific modifications that dramatically extend its activity and potency compared to the naturally occurring hormone.

The two modifications that define IGF-1 LR3:

  • "R3" — an arginine substitution at position 3 of the peptide sequence
  • "Long" — a 13-amino acid N-terminal extension added to the molecule

Together, these changes dramatically reduce IGF-1 LR3's binding affinity to Insulin-like Growth Factor Binding Proteins (IGFBPs) — the proteins that normally sequester circulating IGF-1 in the bloodstream, limiting its availability and duration of action. Natural IGF-1 has a half-life of approximately 12–15 hours; IGF-1 LR3 extends this to 20–30 hours while remaining biologically active for the full period.

The result is a compound with significantly greater anabolic and tissue-repair potency than endogenous IGF-1, making it one of the most extensively studied peptides in muscle physiology, body composition, and regenerative medicine research.

"Exogenous IGF-1 analogs like LR3 and DES offer enhanced bioactivity and specificity, making them useful in both clinical and athletic settings." — Bach, Current Pharmaceutical Design

How It Works — Mechanism of Action

1. IGF-1 Receptor Activation IGF-1 LR3 binds to the IGF-1 receptor (IGF1R), which is expressed on virtually every tissue in the body — with particularly high density on skeletal muscle, tendons, connective tissue, and bone. Receptor activation triggers the PI3K/Akt/mTOR signalling cascade — the primary intracellular pathway driving muscle protein synthesis, cell growth, and tissue repair.

2. mTOR Pathway Activation — Lean Mass Accretion Research mTOR (mechanistic target of rapamycin) is the central regulator of muscle protein synthesis and hypertrophy. IGF-1 LR3's sustained receptor activation drives continuous mTOR signalling across its 20–30 hour half-life, supporting an extended anabolic window that natural IGF-1 cannot maintain.

3. Satellite Cell Activation Satellite cells are muscle stem cells — dormant between training sessions, activated by damage and growth signals. IGF-1 LR3 potently activates satellite cells, increasing their proliferation and fusion into existing muscle fibres. This is the cellular mechanism underlying genuine muscle hypertrophy (new muscle fibre growth) rather than simple intracellular protein accumulation.

4. Glucose Uptake & Nutrient Partitioning Like native IGF-1, LR3 stimulates GLUT4 translocation to cell membranes in muscle tissue, improving glucose uptake independently of insulin. This improves nutrient partitioning — directing carbohydrates preferentially toward muscle tissue rather than adipose storage. This mechanism is also responsible for IGF-1 LR3's hypoglycaemic risk (see side effects).

5. Collagen & Connective Tissue Regeneration IGF-1 receptors are abundant in tendons, ligaments, and cartilage. IGF-1 LR3 stimulates collagen synthesis and fibroblast activity in connective tissue — relevant for injury recovery and joint health alongside its muscle-building effects.

Research-Backed Benefits

Lean Mass Accretion & Hypertrophy Research IGF-1 is the primary downstream mediator of growth hormone's anabolic effects on muscle. LR3's extended half-life and IGFBP resistance make it significantly more potent than native IGF-1 for driving:

  • Muscle protein synthesis
  • Satellite cell proliferation and fusion
  • Lean mass accrual in research models of sarcopenia and muscle wasting

"IGF-1 plays a central role in skeletal muscle hypertrophy through mTOR-dependent and independent pathways." — Clemmons, The Journal of Clinical Endocrinology & Metabolism

Adipose Metabolism & Body Composition IGF-1 LR3 promotes lipolysis (fat breakdown) through GH-like metabolic effects and improves nutrient partitioning — directing energy substrates toward muscle rather than fat storage. Body composition improvements in research settings typically show simultaneous lean mass gains and fat mass reduction.

Connective Tissue Regeneration & Repair Research Strong evidence for IGF-1's role in accelerating recovery from:

  • Muscle tears and strains
  • Tendon and ligament injuries
  • Bone fractures (IGF-1 stimulates osteoblast activity)
  • Post-surgical tissue healing

Research Applications

  • Muscle wasting studies: sarcopenia, cachexia, and age-related muscle loss
  • Growth disorder research: GH deficiency and growth failure models
  • Metabolic research: glucose and lipid metabolism regulation
  • Ageing studies: age-related muscle and bone loss prevention

Dosing & Administration

⚠️ Important Warning — Read Before Dosing: IGF-1 LR3 is one of the most potent anabolic compounds in the research peptide space. Its insulin-mimicking properties create a genuine hypoglycaemia risk. Never inject IGF-1 LR3 in a fasted state. Always have fast-acting carbohydrates immediately available when injecting. Never exceed recommended doses. Medical supervision is strongly advised.

Research Protocol Dose Range: 20–60 mcg per day Route: Subcutaneous injection (systemic effects) Timing: Post-exercise injection is most common — takes advantage of elevated IGF-1 receptor sensitivity in trained muscle tissue

Protocol Guidelines:

ApplicationDoseDuration
Research Protocol40 mcg/day4 weeks, then 2-week break
Body Composition Study60 mcg/day6 weeks maximum
Recovery Research20–40 mcg post-exerciseTraining study duration
Metabolic Research40 mcg/dayWith metabolic monitoring

Critical Dosing Rules:

  1. Start at the lowest dose (20 mcg) and assess tolerance before increasing
  2. Never inject fasted — always have a meal or fast-acting carbohydrates within 30–60 minutes of injection
  3. Refrigerate immediately after reconstitution — use within 7–14 days maximum
  4. Protect from light and temperature fluctuations — IGF-1 LR3 is more sensitive to degradation than most peptides
  5. Keep cycle lengths short — maximum 4–6 weeks with mandatory equal-length breaks between cycles

Reconstitution:

  • Use acetic acid solution (0.1–1%) rather than bacteriostatic water — IGF-1 LR3 reconstitutes more stably in acetic acid
  • Add solution slowly down the vial wall — do not inject directly onto powder
  • Swirl gently — never shake or vortex
  • Refrigerate immediately at 2–8°C
  • Discard if solution is cloudy, discoloured, or contains particles
  • Use within 7–14 days of reconstitution

Side Effects & Safety

IGF-1 LR3 carries a meaningfully higher risk profile than most research peptides due to its potent insulin-mimicking activity and growth-promoting properties. It should be approached with greater caution than other peptides in this catalogue.

Hypoglycaemia (Low Blood Sugar) — Primary Risk IGF-1 LR3 stimulates glucose uptake into muscle cells independently of insulin. At its 20–30 hour half-life, blood glucose can be lowered multiple times throughout the day, not just immediately post-injection.

Symptoms of hypoglycaemia:

  • Dizziness and light-headedness
  • Sweating
  • Shakiness or trembling
  • Brain fog, confusion, or difficulty concentrating
  • Fatigue or sudden energy drop

"IGF-1 administration increases glucose disposal and insulin sensitivity but also increases the risk of hypoglycemia, particularly in fasted states." — LeRoith, Trends in Endocrinology & Metabolism

Management: Always inject with food or fast-acting carbohydrates available. Never inject in a fully fasted state.

Water Retention & Joint Stiffness Mild fluid retention — particularly when stacked with GH or MK-677 — can cause:

  • Puffy hands or feet
  • Morning joint stiffness
  • Mild carpal tunnel-like sensations

Fatigue & Mental Fog Rapid or fluctuating changes in blood glucose can affect energy levels and mental clarity — particularly with inconsistent dosing or low carbohydrate intake.

Serious Long-Term Risks (Extended or Excessive Use)

  • Insulin Resistance — chronic IGF-1 LR3 exposure can paradoxically downregulate insulin receptors and impair glucose metabolism, particularly when stacked with other anabolic compounds or a high-calorie diet
  • Cell Overproliferation — IGF-1 is mitogenic (stimulates cell division). While it does not directly cause cancer, individuals with a personal or family history of hormone-sensitive cancers, or any undiagnosed growths, should avoid use entirely
  • Acromegaly-like Effects — at excessive doses or with very long cycles, IGF-1 LR3 can stimulate growth in soft tissues including organs. Symptoms include joint pain, enlarged hands/feet, and facial changes. This risk is dose and duration dependent.

Who Must Avoid IGF-1 LR3:

  • Anyone with a personal or family history of cancer
  • Individuals with diabetes or impaired glucose regulation
  • Those on insulin therapy
  • Pregnant or breastfeeding women
  • Anyone with active organ or tissue growth disorders
