What Is GHK-Cu?

GHK-Cu is a naturally occurring copper-binding tripeptide — composed of glycine, histidine, and lysine bound to a copper ion — first identified in human plasma in 1973 (Pickart, 1973). It is one of the most extensively studied regenerative molecules in modern biogerontology and wound-healing research, with a scientifically documented ability to activate over 400 human genes involved in tissue repair, collagen synthesis, antioxidant defence, and cellular regeneration (Pickart et al., 2015).

GHK-Cu is naturally present in blood plasma, saliva, and urine, but levels decline significantly with age — dropping from approximately 200 ng/ml in young adults to around 80 ng/ml by age 60 (Pickart, 1973). This age-related decline is believed to contribute to slower wound healing, reduced skin integrity, and declining regenerative capacity. Research supplementation is aimed at restoring these levels to support the body's natural repair systems.

Uniquely among research peptides, GHK-Cu is effective both topically and systemically, making it one of the most versatile compounds in regenerative and cellular ageing research.

How It Works — Mechanism of Action

1. Activates Over 400 Repair Genes

GHK-Cu acts as a broad biological reset signal, modulating gene expression across a remarkable range of repair pathways. Research by Dr. Loren Pickart — who first identified GHK-Cu — demonstrated that it activates genes involved in skin remodelling, extracellular matrix (ECM) construction, fibroblast activity, keratinocyte repair, and angiogenesis, while simultaneously silencing genes associated with inflammation and cancer progression (Pickart et al., 2015; Pickart et al., 2018).

2. Reduces Inflammation

GHK-Cu suppresses key pro-inflammatory cytokines including TNF-α, IL-6, and NF-κB pathway activity (Pickart et al., 2018). This translates to reduced redness, irritation, swelling, and oxidative tissue damage — relevant both for skin health and systemic inflammatory conditions.

3. Stimulates Collagen & Elastin Production

Research demonstrates that GHK-Cu drives dramatic increases in (Maquart et al., 1993; Pickart et al., 2015):

  • Type I and Type III collagen
  • Glycosaminoglycans (GAGs) — the structural molecules in connective tissue
  • Elastin fibres — responsible for skin elasticity and recoil
  • Integrins — cell adhesion proteins critical to tissue remodelling

This mechanism underpins its study in dermal matrix and collagen synthesis research.

4. Enhances Wound Healing

GHK-Cu improves fibroblast migration to wound sites, promotes blood flow and cellular oxygenation, and accelerates tissue remodelling (Maquart et al., 1993). It has been studied in clinical contexts including burns, surgical wounds, scarring, and chronic skin abrasions.

5. Activates Broad Cellular Ageing Signalling

GHK-Cu supports stem cell activation pathways, mitochondrial protection, antioxidant enzyme expression (including superoxide dismutase and catalase), and DNA repair gene expression — giving it a systemic cellular ageing research profile that extends well beyond skin-level effects (Pickart et al., 2018).

⚠️ Note: GHK-Cu injectable formulations are currently classified by Australian regulators as requiring careful consideration due to potential for immune reactions from impurities in unverified sources. Always source from a quality-verified supplier and consult a healthcare professional before commencing any injectable protocol.

Side Effects & Safety

GHK-Cu has an excellent safety profile in the published literature, with no established toxicity at research doses. Its natural presence in the human body and its role as a signalling molecule rather than a pharmacological agent contribute to its favourable tolerability.

Commonly reported (mild)

  • Mild injection site redness or swelling — typically resolves quickly with proper site rotation
  • Rare: mild skin flushing or warmth

Important safety considerations

  • Injectable GHK-Cu quality is highly source-dependent. Impurities in poorly manufactured products have been associated with immune reactions. This is a source quality issue, not an inherent peptide toxicity issue — which reinforces the importance of using reputable, purity-verified suppliers
  • Individuals with copper metabolism disorders (e.g. Wilson's disease) should avoid GHK-Cu supplementation entirely
  • Pregnant or breastfeeding women should avoid use
  • Those with autoimmune conditions should consult a physician — GHK-Cu's immune-modulating properties warrant medical oversight in this population
