What Is GHK (Basic)?

GHK (Glycine-Histidine-Lysine) is a naturally occurring tripeptide first isolated from human plasma in 1973 by Loren Pickart, who identified it as the active factor responsible for the liver's ability to restore its own synthesis capacity after injury. It is found throughout the body — in plasma, saliva, and urine — with plasma concentrations declining significantly with age: from approximately 200 ng/mL at age 20 to around 80 ng/mL by age 60, a 60% reduction that parallels the decline in tissue repair capacity and regenerative ability observed with ageing.

GHK-Cu (copper-bound GHK) is the more widely studied and commercially prevalent form — the copper ion substantially amplifies the peptide's biological activity. GHK (Basic) — the copper-free form offered here — is the parent tripeptide itself, and retains meaningful biological activity through its own receptor binding and gene expression effects, independently of copper chelation.

GHK Basic is particularly relevant for:

  • Applications where copper load is a concern (individuals with Wilson's disease risk or copper sensitivity)
  • Topical skincare formulations where the peptide's gene-regulatory effects are desired without the green pigmentation of GHK-Cu
  • Research contexts investigating the peptide's intrinsic (copper-independent) mechanisms
  • Sensitive skin applications where the oxidative activity of copper is contraindicated

"GHK-Cu promotes skin regeneration by modulating the expression of over 4,000 human genes involved in collagen synthesis, anti-inflammatory signalling, and antioxidant defence." — Pickart & Margolina, Scientific World Journal (2018)

How It Works — Mechanism of Action

1. Gene Expression Modulation — The Primary Mechanism GHK's most remarkable biological property is its ability to modulate the expression of a large proportion of the human genome. Research by Pickart and Margolina (2018) identified that GHK-Cu influences the expression of over 4,000 human genes — approximately 32% of genes associated with biological ageing — resetting them toward a more youthful expression pattern. The GHK tripeptide itself contributes significantly to this gene regulatory activity independently of copper.

Key gene expression effects:

  • Upregulation of collagen I, III, and IV synthesis
  • Upregulation of elastin and proteoglycan production
  • Downregulation of matrix metalloproteinases (MMPs) — enzymes that break down the extracellular matrix
  • Upregulation of antioxidant defence genes (SOD, catalase)
  • Downregulation of pro-inflammatory signalling pathways

2. Fibroblast Activation & Collagen Synthesis GHK directly stimulates fibroblast proliferation and activity — fibroblasts are the primary cells responsible for producing collagen, elastin, and the extracellular matrix proteins that give skin and connective tissue their structure, strength, and elasticity. The result is increased collagen density, improved skin firmness, and enhanced wound healing.

3. TGF-ÎČ Pathway Modulation GHK influences Transforming Growth Factor-ÎČ (TGF-ÎČ) signalling — a primary regulator of fibrosis, tissue repair, and inflammation. It promotes tissue repair-oriented TGF-ÎČ activity while moderating the pro-fibrotic effects that cause excessive scarring.

4. Anti-Inflammatory Activity GHK reduces pro-inflammatory cytokine signalling — particularly TNF-α and IL-6 — through NF-ÎșB pathway modulation. This anti-inflammatory activity contributes to its wound healing and skin restoration effects and supports its role in systemic anti-inflammatory protocols.

5. Angiogenesis Support GHK promotes the formation of new blood vessels (angiogenesis) in healing tissue — ensuring adequate blood supply and nutrient delivery to support the active repair process. This is particularly relevant in wound healing, skin regeneration, and connective tissue repair.

6. Antioxidant Defence GHK upregulates key antioxidant enzymes including superoxide dismutase (SOD) and catalase, reducing oxidative damage in target tissues. This mechanism is particularly valuable in skin ageing applications where ultraviolet-induced oxidative stress is a primary driver of photo-ageing.

Research-Backed Benefits

Dermal Matrix & Collagen Synthesis Research

  • Increased skin thickness, firmness, and elasticity in human clinical studies
  • Reduced fine lines and wrinkles via collagen and elastin upregulation
  • Improved skin hydration and texture
  • Reduced age spots and improved skin clarity
  • Accelerated wound healing with reduced scarring

"GHK-Cu significantly increased skin thickness and improved skin laxity in a double-blind vehicle-controlled clinical trial." — Leyden et al., Skin Pharmacology and Physiology (2008)

Wound Healing & Tissue Repair

  • Accelerated closure of acute wounds in animal and human models
  • Improved healing in chronic wounds and ulcers
  • Reduced excessive scarring (hypertrophic scars and keloids) via TGF-ÎČ modulation
  • Enhanced post-surgical tissue recovery

Hair Follicle Stimulation

  • GHK has demonstrated stimulation of hair follicle growth and improved hair density in preclinical research
  • Activates quiescent hair follicle stem cells
  • May support androgenic alopecia research protocols

Anti-Inflammatory & Connective Tissue

  • Reduces joint inflammation in arthritic models
  • Supports connective tissue integrity in tendons and ligaments
  • Systemic anti-inflammatory effects relevant to chronic inflammatory conditions

Neurological Research

  • Emerging evidence for neuroprotective effects — GHK influences gene expression in brain tissue
  • Potential cognitive and neurological benefits in ageing models under investigation

Dosing & Administration

GHK Basic is primarily used in two ways: topical and injectable.

Topical Application (Primary Use Case for GHK Basic)

  • Add to your existing skincare formulation (serum, cream, moisturiser) at a concentration of 1–5%
  • Apply to clean skin once or twice daily
  • GHK is water-soluble and compatible with most skincare bases
  • Most appropriate for: anti-ageing skincare, wound healing, scar reduction, rosacea and inflammatory skin conditions
  • No green pigmentation (unlike GHK-Cu) — cosmetically clean formulation

Subcutaneous Injection (Systemic Use)

  • Dose: 1–2 mg per injection
  • Frequency: Daily or every other day
  • Cycle: 4–8 weeks on, 2–4 weeks off
  • Sites: Abdomen, upper thigh, or glute — rotate with each injection
  • For systemic anti-inflammatory, connective tissue, and anti-ageing applications

Reconstitution:

  • Add bacteriostatic water to lyophilised powder
  • Swirl gently — never shake
  • Refrigerate at 2–8°C and use within 28 days
  • Discard if solution is cloudy or contains visible particles

Note on GHK vs GHK-Cu: If your primary goals are wound healing, hair growth stimulation, or systemic anti-ageing, GHK-Cu (copper-bound) is generally considered the more potent option due to the copper ion's synergistic biological activity. GHK Basic is preferred when copper is contraindicated, for topical cosmetic applications (no green tint), or for sensitive skin.

Side Effects & Safety

GHK has an excellent safety profile established across decades of cosmetic and clinical research use.

Reported side effects:

  • Mild injection site redness or discomfort — transient, resolves with site rotation
  • Topical: very rare mild skin irritation at high concentrations in sensitive individuals — reduce concentration if observed

Key safety profile:

  • No systemic toxicity reported at therapeutic concentrations
  • No dependency or tolerance
  • Well-tolerated in topical use across all skin types including sensitive skin
  • Substantially better-tolerated topically than GHK-Cu in individuals with copper sensitivity or reactive skin

Important safety considerations:

  • Wilson's disease or copper metabolism disorders — GHK Basic (copper-free) is the appropriate choice; avoid GHK-Cu
  • Pregnancy and breastfeeding — avoid injectable use; topical use at low concentrations is likely safe but consult a healthcare professional
  • Known peptide hypersensitivity — patch test topical formulations before full application
