What Is Adamax?

Adamax is the most potent derivative in the Semax peptide family — a heavily modified synthetic analogue of adrenocorticotropic hormone (ACTH 4-10) that was originally developed by the now-defunct Mexico-based peptide company Ceretropic. While standard Semax was developed by the Institute of Molecular Genetics of the Russian Academy of Sciences in the 1980s, Adamax represents a significant structural evolution beyond both standard Semax and N-Acetyl Semax Amidate, engineered for maximum potency, blood-brain barrier penetration, and duration of action.

Structurally, Adamax is an N-acetyl Semax base that has been modified at the C-terminus with the adamantyl portion of P21 (Peptide 021). This means it carries two critical modifications:

  • N-terminus: An acetyl group (CH₃CO-) — the same modification found in N-Acetyl Semax, which increases stability and improves blood-brain barrier penetration by enhancing lipophilicity.
  • C-terminus: An adamantane group — a large, rigid, lipophilic diamondoid structure borrowed from P21 (a CNTF-derived peptide). This is the key differentiator: while N-Acetyl Semax Amidate uses a simple amide group at the C-terminus, Adamax uses the substantially more advanced adamantane moiety.

The adamantane group is what sets Adamax apart from all other Semax derivatives. It provides three critical advantages over the amide C-terminus used in N-Acetyl Semax Amidate:

  1. Superior enzymatic protection — the rigid cage structure physically shields the C-terminus from carboxypeptidase degradation more effectively than a simple amide bond.
  2. Thermal stability — the adamantane moiety buffers against temperature-induced breakdown, which amide groups cannot do. This has practical implications for storage and transport stability.
  3. Dramatically enhanced BBB penetration — the lipophilic nature of the adamantane group allows Adamax to cross the blood-brain barrier far more efficiently than N-Acetyl Semax Amidate, resulting in higher CNS bioavailability and more sustained neurological effects.

Molecular formula: C₂₂H₅₂N₁₆O₆

"Adamax is an N-acetyl semax base limited at the C-terminus with the adamantyl portion of P21. This modification makes Adamax more powerful and efficient than its predecessors." — Ceretropic (original manufacturer documentation)

How It Works — Mechanism of Action

1. Superior Blood-Brain Barrier Penetration via Adamantane The adamantane group is a highly lipophilic tricyclic alkane that dramatically increases the peptide's ability to cross the blood-brain barrier. The rigid cage moiety protects nearby functional groups from metabolic cleavage, enhances stability and distribution in blood plasma, and allows the compound to reach target brain tissues at higher concentrations than any other Semax derivative. This is the primary structural advantage of Adamax over N-Acetyl Semax Amidate.

2. BDNF Upregulation & TrkB Receptor Sensitisation Adamax's defining neurological effect is potent upregulation of Brain-Derived Neurotrophic Factor (BDNF) in the hippocampus and prefrontal cortex. Crucially, Adamax also increases the sensitivity of TrkB receptors — the primary signalling receptors for BDNF. By making TrkB receptors more responsive to BDNF, Adamax enhances neuroplasticity, synaptic formation, and long-term potentiation to a greater degree than standard Semax or N-Acetyl Semax Amidate.

3. Dopamine, Norepinephrine & Serotonin Modulation Adamax modulates dopaminergic, noradrenergic, and serotonergic neurotransmission — enhancing the sensitivity and efficiency of these pathways rather than flooding them with exogenous agonists. This produces improved motivation, mood stability, mental drive, and sustained focus without the tolerance, dependency, or post-dose crash associated with conventional stimulants. Anecdotal reports describe stimulant-like effects lasting 8–10 hours without overstimulation.

4. Enhanced Stability via Dual Terminal Protection The N-terminal acetyl group renders the peptide resistant to leucine aminopeptidase degradation, while the C-terminal adamantane provides superior protection against carboxypeptidase enzymes — exceeding the protection offered by the amide group in N-Acetyl Semax Amidate. The compound survives significantly longer in both plasma and brain tissue, translating to more sustained neurological effects from a single dose.

5. Neuroprotection via Anti-Inflammatory Pathways Adamax downregulates pro-inflammatory cytokines in brain tissue, activates anti-apoptotic signalling in neurons, and protects against oxidative stress and neuronal death. These neuroprotective properties underlie its emerging research interest in neurodegenerative disease models, particularly Alzheimer's disease.

6. Neurogenesis Promotion Adamax promotes the formation of new neurons (neurogenesis) in the hippocampus — the brain region central to learning, memory, and emotional regulation. This suggests potential for brain tissue regeneration and repair, extending beyond simple neuroprotection into active neurological restoration.

7. Melanocortin Receptor Activity As an ACTH 4-10 analogue, Adamax retains partial melanocortin receptor (MC4R) activity — a receptor family implicated in cognitive processing, attention, and memory consolidation. This contributes to nootropic effects independently of the BDNF pathway.

Dosing & Administration

⚠️ For research purposes only. Not for human therapeutic use. All dosing information is derived from published research literature and community-reported protocols, and is provided for educational reference only. Adamax is an experimental research compound with no published human clinical trials.

Available Routes: Subcutaneous Injection & Intranasal

Adamax is significantly more potent than standard Semax and N-Acetyl Semax Amidate. Doses are correspondingly lower. Due to its experimental status, all dosing should begin at the conservative end of the range.

Sub-Q Administration Protocol

ProtocolDoseFrequencyCycle
Conservative (beginner)100–300 mcgOnce daily4–8 weeks on, 2–4 weeks off
Standard research300–500 mcgOnce daily8–12 weeks on, 2–4 weeks off
Advanced protocol500 mcg–1 mgOnce daily8–12 weeks on, 4 weeks off

Intranasal Administration Protocol

ProtocolDoseFrequencyCycle
Conservative (beginner)100–200 mcg1–2 sprays daily4–8 weeks on, 2–4 weeks off
Standard research200–400 mcg1–2 sprays daily8–12 weeks on, 2–4 weeks off

Note: Intranasal dosing based on Ceretropic's original recommendation of 100–200 mcg per spray from a 10mg/10mL nasal solution.

Injection Technique (Sub-Q):

  • Inject subcutaneously into the abdomen (preferred) or outer thigh — pinch the skin and insert at 45°
  • Rotate injection sites across cycles to prevent localised irritation
  • Allow the reconstituted solution to reach room temperature before injection

Timing

  • Morning dosing strongly preferred — the compound's activating/dopaminergic properties can interfere with sleep onset if administered in the afternoon or evening
  • Effects typically begin within 30–60 minutes and are sustained for 8–10 hours due to the extended half-life provided by the adamantane modification
  • Some users report 300 mcg as too potent for their first cycle — start low and titrate up

Cycle Rationale Cycling is essential to preserve receptor sensitivity and avoid downregulation of BDNF and melanocortin pathways. The 4–8 week on / 2–4 week off structure is based on community protocols. Do not exceed 12 consecutive weeks at maximum doses. Given that Adamax has no published human clinical data, conservative cycling is strongly recommended.

Storage

  • Lyophilised (unreconstituted): Store at 2–8°C. Stable for up to 24 months when sealed and refrigerated. The adamantane modification provides additional thermal stability compared to other Semax derivatives, but refrigeration remains the recommended storage condition.
  • Reconstituted: Refrigerate at 2–8°C. Use within 28 days. Do not freeze reconstituted solution. Keep away from light.

Side Effects & Safety

Adamax shares the generally favourable safety profile of the Semax compound class, but with an important caveat: Adamax has no published human clinical trials and no peer-reviewed Adamax-specific research. All safety information is derived from the parent compound (Semax) safety data and community-reported anecdotal experiences. The founding peptide chemist at Ceretropic explicitly stated: "This is a brand new peptide we developed with no human clinical research, or any research of any kind. It should be treated as a research compound."

Commonly reported (mild and transient):

  • Mild injection site irritation — typical of sub-Q peptide administration
  • Headache — particularly at initiation or with higher doses
  • Insomnia or sleep disturbance — particularly if dosed in the afternoon or evening
  • Mild anxiety or restlessness — more common at doses above 500 mcg
  • Nasal irritation (intranasal route only)

Less commonly reported:

  • Elevated blood pressure
  • Heart palpitations
  • Nausea, vomiting, or diarrhoea
  • Mood changes or emotional lability (rare)

Important Safety Notes:

  • Adamax is significantly more potent than standard Semax and N-Acetyl Semax Amidate — doses should be scaled accordingly
  • Due to the lack of clinical data, unknown long-term effects cannot be ruled out
  • Individuals with cardiovascular conditions should exercise caution due to potential blood pressure effects
  • Do not combine with MAOIs or SSRIs without medical supervision
  • Do not combine with other stimulants without careful consideration of cumulative effects
  • Discontinue use and consult a healthcare professional if any adverse reactions occur
