What Is CJC/IPA?

CJC/IPA is a pre-mixed, lyophilised blend of two complementary growth hormone secretagogues — CJC-1295 (no DAC) and Ipamorelin — combined into a single 10mg vial (5mg CJC-1295 / 5mg Ipamorelin, 1:1 ratio). It is one of the most widely used growth hormone peptide combinations in research settings, valued because the two components trigger GH release through completely different receptor systems, producing a combined effect substantially greater than either compound alone.

You may also see this blend written as CJC IPA, CJC + IPA, CJC & IPA or CJC and IPA blend — they all refer to the same CJC-1295 (no DAC) and Ipamorelin combination. Researching the two compounds separately? See CJC-1295 No DAC and Ipamorelin.

CJC-1295 (also known as Mod GRF 1-29) is a synthetic analogue of growth hormone-releasing hormone (GHRH). It mimics the hypothalamus's natural signal to the pituitary gland, binding GHRH receptors on pituitary somatotroph cells with roughly four times the binding affinity of the native hormone (in vitro receptor binding study, 2010). This is the "accelerator" — it tells the pituitary to produce and release growth hormone.

Ipamorelin is a pentapeptide (five amino acids, 711.85 Da) that works through an entirely separate pathway — the ghrelin receptor (GHSR1a). When it binds this receptor on pituitary somatotrophs, it triggers a calcium-dependent GH release that amplifies whatever GHRH signalling is already occurring. A cellular study (Cunha & Mayo, 2002) demonstrated that active ghrelin receptor signalling potentiates GHRH signalling — the two pathways don't simply add together, they multiply.

Unlike older GH secretagogues such as GHRP-2 and GHRP-6, Ipamorelin is highly selective for the GH pathway. Raun et al. (1998) showed zero significant cortisol or ACTH elevation even at 200 times the effective GH-releasing dose — a finding that established Ipamorelin as the preferred GHRP for clean, predictable protocols.

This is a pre-mixed lyophilised powder blend — not a liquid. As with all Riptide Health lyophilised products, it requires reconstitution with bacteriostatic water before use.

How the Stack Works — Dual-Pathway Mechanism

CJC-1295: The Signal Sender

CJC-1295 without DAC is a 30-amino-acid GHRH analogue weighing 3,367.97 Daltons, with a half-life between 30 minutes and 2 hours. That short half-life is the point — natural GH secretion is pulsatile, with the largest pulse occurring during deep sleep, and CJC-1295 without DAC preserves that natural rhythm.

  • A human study (24 subjects, 100mcg three times daily for 30 days, 2008) confirmed preserved natural GH pulsatility with no pituitary desensitisation.
  • A pharmacokinetic study (12 subjects, 2006) showed peak GH at 15–30 minutes post-injection with doses of 100–200mcg.

Ipamorelin: The Clean Releaser

Ipamorelin's selectivity is what separates it from older GH secretagogues. All ghrelin receptor agonists (GHRP-2, GHRP-6, Ipamorelin) bind the same receptor, but GHRP-2 and GHRP-6 also elevate cortisol, prolactin, and appetite. Ipamorelin does not.

  • Raun et al. (1998): zero significant cortisol or ACTH elevation even at 200x the effective GH-releasing dose.
  • A GH stimulation study (2019) found consistent GH elevation 30–60 minutes post-injection at 200mcg daily over 8 weeks.
  • A body composition trial (2020, 250mcg daily, 12 weeks) documented lean mass gains and bone density improvements in adults over 45.
  • A sleep study (2021) showed improved sleep latency and increased slow-wave sleep at 200mcg before bed.

Combined Effect

CJC-1295 signals the pituitary to produce GH. Ipamorelin signals the pituitary to release it. Two independent signals, two receptor pathways, converging on the same somatotroph cells to create one amplified GH pulse — followed by elevated IGF-1, supporting muscle protein synthesis, fat metabolism, and tissue recovery.

Relative GH pulse magnitude (approximate, based on published data):

ProtocolRelative GH Pulse
Ipamorelin alone3x baseline
CJC-1295 (DAC, sustained)3x baseline
CJC-1295 (no DAC) alone4x baseline
CJC-1295 + Ipamorelin8x baseline

Timing Rules

GH release is blunted by insulin and elevated blood glucose, so timing matters more for this stack than for most peptides.

  • Inject on an empty stomach, at least 2–3 hours after your last meal
  • Wait 20–30 minutes after injection before eating
  • Before bed is the optimal timing — it amplifies the natural nocturnal GH surge
  • If dosing twice daily, morning (fasted) and bedtime are the standard windows
  • Post-workout is an option for a third daily dose, but only if you haven't eaten recently

Cycle Structure

  • On cycle: 8–16 weeks
  • Off cycle: 4–8 weeks
  • Why cycle: Ipamorelin's receptor desensitisation risk increases after 3–4 months of continuous use (2023 safety analysis). Cycling off resets sensitivity.
  • Monitor: IGF-1 bloodwork at baseline, mid-cycle (week 6–8), and 4 weeks post-cycle

Reconstitution

  • Add bacteriostatic water slowly down the vial wall — do not inject directly onto the powder
  • Swirl gently — never shake
  • Refrigerate at 2–8°C and use within 28 days
  • Discard if solution is cloudy or contains visible particles

⚠️ Important: Consult a qualified healthcare professional before commencing use, particularly if you have a history of cancer, diabetes or insulin resistance, or carpal tunnel/nerve compression issues.

How It Compares to Alternatives

vs Sermorelin: Sermorelin is the original GHRH analogue, FDA-approved in 1997 (discontinued 2008 for manufacturing reasons, not safety). Its half-life is only 10–12 minutes with 6% subcutaneous bioavailability, compared to CJC-1295's 30-minute to 2-hour half-life and higher receptor affinity. Sermorelin has the strongest human evidence base of any GHRH analogue — Corpas et al. (1992) doubled 12-hour GH release in 60–78 year old males over 6 weeks — and is synergistic with Ipamorelin (3–5 fold GH increases documented). Sermorelin wins on evidence; CJC-1295 wins on pharmacokinetics.

vs GHRP-2: GHRP-2 is more potent than Ipamorelin at raw GH stimulation — combined with a GHRH peptide, GHRP-2 has produced up to 54-fold GH increases vs 20-fold with GHRH alone (Domestic Animal Endocrinology, 2000). But this comes with trade-offs: increased cortisol at higher doses, prolactin elevation, and a 35.9% increase in food intake (JCEM, 2010). Ipamorelin's selectivity avoids these side effects.

vs MK-677 (Ibutamoren): MK-677 is an oral ghrelin mimetic — no injections required, but it provides continuous GH elevation similar to CJC-1295 DAC, raising desensitisation concerns, and significantly increases appetite with insulin resistance risk on extended use. CJC-1295 + Ipamorelin offers more precise control over GH timing and magnitude.

Side Effects & Safety

CJC-1295 + Ipamorelin has a reputation as one of the milder GH secretagogue protocols. That reputation is mostly deserved, with a few caveats.

Commonly reported (generally mild and transient)

  • Facial flushing and warmth, 5–10 minutes post-injection — resolves on its own; more common with CJC-1295 than Ipamorelin
  • Mild water retention in the first 1–2 weeks, usually stabilises
  • Vivid dreams and improved sleep — a side effect most users consider a benefit, caused by GH's effect on slow-wave sleep
  • Mild hunger increase 20–30 minutes post-injection from Ipamorelin's ghrelin receptor activity — much less pronounced than with GHRP-6
  • Injection site reactions — minor redness or irritation, standard for subcutaneous peptides

Serious concerns

  • Immunogenicity risk has been flagged for both peptides, including rare anaphylactic reactions
  • Cardiovascular: transient flushing, warmth, and hypotension post-injection
  • Carpal tunnel symptoms (numbness, tingling in hands) signal excessive GH/IGF-1 — dose should be reduced if these occur
  • GH antagonises insulin — those with diabetes or insulin resistance need to monitor blood glucose closely
  • Persistent joint pain beyond the first 2 weeks is a sign to reduce dose or pause

⚠️ Cancer Contraindication: Growth hormone promotes cell proliferation. Anyone with active cancer, a history of cancer, or unscreened tumours should not use GH secretagogues. This applies to CJC-1295, Ipamorelin, and all GH-axis compounds alike.

Contraindications

  • Active malignancy or cancer history
  • Diabetic retinopathy
  • Severe kidney disease
  • Pregnancy or breastfeeding
  • Existing carpal tunnel or nerve compression
  • Pituitary tumours

Monitoring

  • IGF-1 levels: baseline, mid-cycle (week 6–8), and 4 weeks post-cycle
  • Blood glucose: essential for anyone with metabolic concerns
  • Joint symptoms: persistent pain or carpal tunnel signs warrant dose reduction
  • Thyroid function: similar GHRH-analogue studies found a minority of users developed hypothyroidism; monitoring is a reasonable precaution

Synergy Stacks

Foundational GH-Axis Stack: CJC/IPA (standalone)

The dual-pathway mechanism (GHRH + ghrelin receptor) already provides synergy within a single product — this is a complete GH secretagogue protocol on its own.

Recovery & Repair Stack: CJC/IPA + BPC-157 + TB-500

CJC/IPA supports the systemic GH/IGF-1 axis for tissue repair and recovery; BPC-157 and TB-500 provide targeted local tissue-healing signalling. A comprehensive recovery protocol combining systemic and localised repair mechanisms.

Cellular Longevity Stack: CJC/IPA + NAD+ + Epitalon

CJC/IPA addresses the GH-axis dimension of ageing; NAD+ restores cellular energy and sirtuin function; Epitalon targets telomere biology. A multi-pathway approach to cellular ageing research.

Metabolic Research Stack: CJC/IPA + Retatrutide

Retatrutide is a triple GLP-1 / GIP / glucagon receptor agonist — the most advanced compound in the GLP-1 research class, ahead of single-agonist semaglutide and dual-agonist tirzepatide. Pairing it with CJC/IPA combines incretin-pathway metabolic research with GH-axis research. ⚠️ Note: Combining a GH secretagogue with a GLP-1 agonist requires careful blood glucose monitoring given GH's insulin-antagonising effects — medical supervision recommended.
